This study investigated the actions of y-aminobutyric acid (GABA) on mouse ileum and the influence of electrical stimulation on contractions induced by GABA. The mouse ileum was found to be similar to the guinea-pig ileum with respect to the actions of GABA and other agonists such as 5-hydroxytryptamine, nicotine and acetylcholine. GABA, when applied to the mouse ileum, induced a transient contraction through neuronal GABA A receptor activation, followed in some cases by a prolonged relaxation mediated via the GABA B receptor. Electrical field stimulation had an inhibitory effect on contractions induced by GABA and muscimol, a GABA A receptor agonist, but had no effect on contractions induced by other agonists. Several antagonists (naloxone, phenoxybenzamine, propranolol, indomethacin, methysergide, ascorbic acid, theophylline) were unable to prevent this inhibitory effect on GABA-induced contractions. The inhibitory effect was reversed by washing the ileum or allowing recovery periods after stimulation. Greater inhibition was observed when the ileum was stimulated at higher frequencies or for longer periods thus suggesting the release of an inhibitory substance upon stimulation. The inhibitory effect could not be transferred via physiological solution from a stimulated donor ileum to an unstimulated recipient ileum, perhaps due to rapid destruction of the inhibitory substance. Capsaicin selectively diminished contractions induced by GABA, suggesting that this compound may release an endogenous inhibitory peptide substance similar to that released upon electrical stimulation. Availability of specific antagonists for endogenous inhibitory peptides that are released upon electrical stimulation would help to resolve the nature of this putative inhibitory substance.
