"Early detection of illness is critical both for controlling the spread of disease and for administering successful treatment, but it requires rapid and sensitive diagnostic capabilities. Label-free sensing provides a significant advantage in achieving this goal because it eliminates the need for auxiliary reagent materials or sample preparation. Various refractive index (RI) based optical sensors have shown promise as sensitive, label-free platforms for pathogen detection. Sensing platforms using two-dimensional photonic bandgap crystal structures, in particular, have shown the potential to achieve single-pathogen detection capabilities. The work contained in this dissertation describes the design, fabrication, and characterization of two-dimensional photonic crystal (PhC) structures to be implemented as sensitive, label-free diagnostic sensors for the detection of viral pathogens at low, or even single, copy numbers. Photonic crystals are composite materials in which unique optical properties result from a structurally periodic arrangement of materials with differing refractive indices (RI). Two-dimensional slab-PhCs are one such implementation in which a sub-wavelength layer of high-RI dielectric material is perforated with low-RI holes. By carefully designing the geometry of the slab-PhC, resonant optical cavities that are extraordinarily sensitive to small changes in local RI can be engineered so that infiltration of pathogenic analyte within the PhC structure causes a shift in optical resonance. Reduced optical transmission at the frequency of resonance allows detection to be observed by monitoring the transmission spectrum of the PhC device. These sensor transducers are compatible with standard semiconductor fabrication procedures and with microfluidic channel integration, permitting implementation in a lab-on-a-chip style sensing tool. We have pursued two 2D waveguide-coupled slab-PhC sensor geometries in this regard: one implementing a "small-defect" alteration of a periodic structure array, and the other incorporating a "large-defect" lattice disruption. Both have been interfaced with microfluidic flow channels to perform real-time sensing measurements. Detection of antibody proteins and virus-sized particles was demonstrated with the developed sensing platforms, and single-particle detection capabilities were confirmed for the large-defect PhC sensor. Computational electromagnetic field simulations indicate that both virus capture and small-molecule binding can be simultaneously measured and discerned from one another using this large-defect design, thus enabling background-corrected, single-pathogen target detection"--Pages iii-iv.

The second volume of the book concerns the characterization approach of photonic crystals, photonic crystal lasers, photonic crystal waveguides and plasmonics including the introduction of innovative systems and materials.
